Analysis of Mutation Frequency at hprt Gene from the Mice Exposed to Penfachlorophenol



Abstract
he mutation frequency (MF) at hprt gene was determined in splenic T-cell from mice exposed to pentachlorophenol(PCP) in vivo,. Mice were grown for 1 and 8 months under optional condition fed with food supplements with PCP(0~500ppm). Spleens and
blood
were isolated and both cell types were stimulated with concanavalin A and interleukin 2 and then seeded in medium containing 6-thioguanine to select for hprt negative mutants.
Mutation frequency was statistically significant increase by net dose of treated PCP. The relationship between the net dose of PCP and the mutant frequency in hprt gene was approximately linear over the range of 0~8.96mg of PCP with an linear
regression
equation of [mutant fraction={0.2477x(net) dose of PCP, mg}+0.5940]x10E-6] and [mutation frequency={1.9661x(net dose of PCP, mg)+4.7141}x10E-6].
To mouse model for somatic mutation at the hprt gene, factors of cultivation were tested and discussed.
